The Amaxa nucleofector is optimized to efficiently electroporate hard-to-transfect primary cell types and cell lines. Nucleofection is an alternative to retro/lenti-viral vector-mediated DNA delivery to cell types of interest. Amaxa has developed a multitude of cell type-specific transfection protocols and kits for a total of 608 primary cell types and cell lines. More information on the Amaxa nucleofector can be found here.

We have used the electroporator to transfect CD34+ human hematopoietic stem cells.
Figure 1: Percent GFP+ and viability assayed by 7-AAD exclusion in freshly-thawed CD34+ cells treated with Amaxa’s Nucleofector U-001 program with addition of pGFP control plasmid (A), cells treated with U-001 without plasmid (B) and cells not electroporated (C), 24 hours after electroporation. Increased cell death in electroporated cells was due to both electroporation and thawing.
